State Route 6 is part of Maine's system of numbered state highways, running from the Quebec border in Sandy Bay Township, to the New Brunswick border in Vanceboro. At just over 207 miles (333 km) in length, it is the third-longest state highway in Maine (behind State Route 11 and State Route 9) and its fifth-longest numbered highway overall (U.S. Route 1 and Interstate 95's lengths are much longer). SR 6 is the only highway in the state to both begin and end at the Canada–United States border. Much of the route, particularly towards its western and extreme eastern ends, runs through very isolated parts of the state.
Approximately 70% of the length of SR 6 is concurrent with other highways, most notably U.S. Route 201, SR 15, SR 16 and U.S. Route 2 in Maine. The only section of SR 6 not concurrent with another route is from its junction with US 2 in Lincoln to the New Brunswick border, a distance of 62 miles. Except for this section, locals generally refer to the roads by their other numbers.
